Organoids grown from stem cells are powerful research tools, and researchers can enhance their usefulness in development and disease modeling by introducing genetic mutations. However, using traditional methods of genetic manipulation in organoids is a challenge. To successfully grow desired cell types and support organoid growth and selection after gene editing, researchers must carefully formulate conditioned media. Furthermore, scientists can prepare organoid media that acts as a selection marker after CRISPR/Cas9 editing.

This Technique Talk will teach the tips and tricks needed for growing organoids from stem cells in 3D culture conditions and how to use conditioned media for selection after CRISPR editing.
